Thrombin Calibrator

Thrombin Calibrator should be used in each experiment in every group. After adding 1 mL deionized water to the vial, the solution can be used in the CAT assay. Use 20 uL of Thrombin Calibrator together with 80 uL of plasma in the wells. Based on the calibrated amount of thrombin activity, the concentration of thrombin in time can be calculated from the signal derived from the samples in which thrombin generation takes place. The Thrombin Calibrator is used for each individual plasma to correct for donor-to-donor differences in color of plasma, inner filter effect and substrate consumption.

PPP-Reagent

CAT# 86193 (box of 20 vials * 1mL)

PPP-Reagent is used as a trigger that is added to platelet-poor plasma to initiate thrombin generation.

PPP-Reagent contains a mixture of phospholipids and Tissue Factor. After reconstitution the solution can be used in the assay. Use 20 uL of PPP-Reagent together with 80 uL of platelet-poor plasma in the wells. After addition of 20 uL FluCa the reaction starts.

PPP-Reagent LOW

CAT# 86194 (box of 20 vials * 1mL)

PPP-Reagent LOW is used as a trigger that is added to platelet-poor plasma to initiate thrombin generation.

PPP-Reagent LOW contains a mixture of phospholipids and Tissue Factor. After reconstitution the solution can be used in the assay. Use 20 uL of PPP-Reagent LOW together with 80 uL of platelet-poor plasma in the wells. After addition of 20 uL FluCa the reaction will start.
PPP-Reagent LOW contains a very low concentration of Tissue Factor. This makes it possible to use PPP-Reagent LOW in plasma of hemophilia patients for which a higher concentration of Tissue Factor would make the assay too insensitive to detect the hemophilia. PPP-Reagent LOW is particularly recommended to be used in hemophiliac plasma in order to increase sensitivity to factors VIII, IX and XI.

PPP-Reagent HIGH

CAT# 86195 (box of 20 vials * 1mL)

PPP-Reagent HIGH is used as a trigger that is added to platelet-poor plasma to initiate thrombin generation.

PPP-Reagent HIGH contains a mixture of phospholipids and Tissue Factor. After reconstitution the solution can be used in the assay. Use 20 uL of PPP-Reagent HIGH together with 80 uL of platelet-poor plasma in the wells. After addition of 20 uL FluCa the reaction will start.
PPP-Reagent HIGH contains a higher concentration of Tissue Factor. This makes it possible to use PPP-Reagent HIGH in plasma that contains so much anticoagulant that thrombin generation measurement is difficult, for example in plasma containing unfractionated heparin (UFH). PPP-Reagent HIGH is particularly recommended to be used in plasma containing anti-thrombotics.

PRP-Reagent

CAT# 86196 (box of 20 vials * 1mL)

PRP-Reagent is used as a trigger that is added to platelet-rich plasma to initiate thrombin generation.

PRP-Reagent contains Tissue Factor and a minimal amount of phospholipids. After reconstitution the solution can be directly used in the assay. Use 20 uL of PRP-Reagent together with 80 uL of platelet-rich plasma in the wells. After addition of 20 uL FluCa the reaction will start. PRP-Reagent is used in platetet-rich plasma but can also be used in platelet-poor plasma to look for the existence and activity of microparticles. 
 

FluCa-Kit

CAT# 86197 (box of 20 vials * 1.6mL Fluo-Buffer and 1 vial * 0.9mL Fluo-Substrate)

Fluo-Buffer

Fluo-Buffer together with the appropriate amount of Fluo-Substrate is called FluCa and is used in each experiment in every well. It is dispensed by the machine after which the citrated plasma will be recalcified so that the reaction will start. Fluo-Buffer contains Hepes (pH 7.35) and Calcium Chloride. It comes in vials with screw cap which should be placed in the water bath (37 C) prior to the experiment.

Fluo-Substrate

Fluo-Substrate contains the fluorogenic substrate solubilized in DMSO. Fluo-Substrate is added to the warmed Fluo-Buffer shortly before the experiment.

MP-Reagent

CAT# 86222 (box of 20 vials * 1mL)

MP-Reagent is used as a trigger that is added to platelet-poor plasma to initiate thrombin generation and detect microparticles.

MP-Reagent contains phospholipids only. After reconstitution the solution can be directly used in the assay. Use 20 uL of MP-Reagent together with 80 uL of platelet-poor plasma in the wells. After addition of 20 uL FluCa the reaction will start. Tissue Factor bearing microparticles are able to trigger thrombin generation in plasma. The use of MP-Reagent makes the assay specifically sensitive to Tissue Factor present in circulation on the surface of microparticles.
